EP 2.2.3 Potentiometric Determination of pH
Summary
Ph. Eur. 2.2.3 Potentiometric determination of pH is the official European Pharmacopoeia method used when a monograph requires pH testing. It specifies the principle (glass indicator electrode + reference electrode), apparatus performance, calibration, and measurement conditions to obtain accurate, reproducible pH values. While USP <791>, JP, and Ph. Eur. chapters on pH are conceptually aligned, Ph. Eur. 2.2.3 is not a PDG-harmonized text and is therefore not interchangeable.
Key Principles & Scope
- Applicability. Use whenever a Ph. Eur. monograph calls for pH. Measurements are normally performed at the same temperature used for calibration (typically 20–25 °C), and the temperature of the test solution must be within 2 °C of the buffers unless a valid temperature correction is applied.
- Principle. Measure the potential difference between a hydrogen-ion-sensitive (glass) electrode and a reference electrode; convert the potential to pH via calibration against standard buffers.
- Apparatus. A high-impedance meter with a display discrimination of 0.05 pH unit or better and 1 mV or better. Proper electrode care and conditioning are required. (Combination electrodes are acceptable.)
- Calibration. Calibrate with at least two certified buffer solutions whose pH values bracket the expected sample pH and differ from each other by at least 2 pH units. After calibration, the reading of each standard buffer must not differ from its certified value by more than 0.05 pH units.
- (Note: While a specific electrode slope, such as 95–105% of theoretical, is a common system suitability requirement in many laboratories, it is not a requirement defined within this chapter.)
- Temperature. Follow the chapter’s temperature guidance or apply instrument temperature correction. For highest accuracy, use the same temperature (within 2 °C) for calibration and measurement.
- Non-aqueous/partly non-aqueous systems. If the meter is standardized with aqueous buffers, pH readings of non-aqueous or partially non-aqueous samples are considered approximate.
- Buffers. Reference buffers for calibration may be prepared per Ph. Eur. 4.1.3 Buffer solutions or purchased as certified reference materials traceable to primary standards.
- Measurement essentials. After calibration, rinse electrodes, immerse in the test solution (gentle stirring), and record the pH after stabilization under the same conditions used for the buffers.
