ICH Q8(R2) Pharmaceutical Development
Summary
ICH Q8(R2) sets out harmonized principles for the pharmaceutical development section of regulatory submissions, with an emphasis on Quality by Design (QbD) — a systematic, science- and risk-based approach to developing drug products and manufacturing processes.
It replaces the older “test quality in” mindset with “build quality in” from the outset by linking material attributes and process parameters directly to critical product characteristics.
Key Principles & Scope
Scope
-
Primarily applies to drug product development; principles may also apply to drug substance development, but Q11 is the primary reference for APIs
-
Intended for Module 3.2.P.2 of the Common Technical Document (CTD).
-
The principles are applicable across dosage forms, but implementation is flexible — companies may follow a traditional or enhanced (QbD) approach. QbD is encouraged. Quality cannot be tested into products; it must be built-in by design.
QbD involves starting with predefined objectives and using scientific understanding and risk management to develop a robust manufacturing process.
Key Elements of the QbD Approach
-
Quality Target Product Profile (QTPP)
-
Prospective summary of product characteristics linked to safety, efficacy, manufacturability, and patient use.
-
-
Critical Quality Attributes (CQAs)
-
Physical, chemical, biological, or microbiological properties that must be controlled to meet the QTPP.
-
-
Linking Inputs to Outputs
-
Critical Material Attributes (CMAs) — input material properties affecting CQAs.
-
Critical Process Parameters (CPPs) — manufacturing parameters affecting CQAs.
-
Identified using risk management tools (ICH Q9).
-
-
Design Space (optional but beneficial)
-
Multidimensional range of CMAs and CPPs shown to assure quality.
-
Operating within an approved design space is not considered a regulatory change once the design space is formally reviewed and approved by the regulatory authority
-
-
Control Strategy
-
Planned set of controls on materials, processes, and product testing implemented and refined throughout the product lifecycle, consistent with ICH Q10/Q12 principles.
-
Can enable innovations such as Real-Time Release Testing (RTRT).
-
Relationship to Other ICH Guidelines
-
Q9(R1): Quality Risk Management — provides the risk tools for QbD.
-
Q10: Pharmaceutical Quality System — lifecycle implementation.
-
Q11: Drug Substance development — applies QbD principles to APIs.
-
Q12: Lifecycle management — builds on the flexibility of Design Space.
Regulatory Status
Originally finalized in 2005, revised to R1 in 2008, and to R2 in 2009.
