ICH Q2(R2) Validation of Analytical Procedures
Summary
ICH Q2(R2) is the modernized harmonized guideline for validating analytical procedures, finalized in parallel with its companion guideline, ICH Q14.
The guideline defines the principles and evidence required to demonstrate that an analytical procedure is fit for its intended purpose—capable of consistently delivering accurate, reliable, and precise results
Key Principles & Scope
Primarily applies to commercial QC testing (release, stability) for drug substances/products; in-process, excipient, and exploratory methods are out of scope
Core Validation Characteristics
Assessed depending on the method’s intended use:
-
Specificity – Selective measurement in the presence of other components
-
Accuracy – Closeness to the true value
-
Precision – Repeatability, Intermediate Precision, and, if applicable, Reproducibility
-
Detection Limit (LOD)
-
Quantitation Limit (LOQ)
-
Linearity
-
Range
-
Robustness – Resistance to small, deliberate parameter changes
Modernization for New Technologies
Provides guidance for validating multivariate model-based methods (e.g., NIR, Raman) and hyphenated techniques (e.g., LC–MS), including model maintenance, periodic verification, and revalidation when models are updated.Lifecycle Perspective
Validation is part of a continuous lifecycle, linking initial validation with ongoing performance verification and continual improvement under ICH Q10.Synergy with ICH Q14
-
Q14: Method development and scientific understanding
-
Q2(R2): Formal validation of the chosen procedure
Enhanced Approach data from Q14 (e.g., Design of Experiments robustness studies) can directly support Q2(R2) requirements.
