ICH Q4B ANNEX 14

ICH Q4B Annex 14 formally declares that the Bacterial Endotoxins Test (BET) procedures in the European Pharmacopoeia (EP), Japanese Pharmacopoeia (JP), and United States Pharmacopeia (USP) are scientifically equivalent and interchangeable in the ICH regions. This conclusion, reached through the ICH Q4B evaluation process, means a manufacturer can follow any one of these compendial chapters in full and have the results accepted by regulatory authorities in Europe, Japan, and the USA without re-testing or re-validation, provided the chapter is adopted in its entirety; selective mixing of methods or conditions from different chapters is not permitted.

Annex 14 covers the following harmonized pharmacopoeial chapters:

• EP: Chapter 2.6.14 – Bacterial Endotoxins

• JP: Chapter 4.01 – Bacterial Endotoxins Test

• USP: General Chapter <85> – Bacterial Endotoxins Test

All three chapters include the officially recognized BET methods including gel-clot, kinetic-turbidimetric, kinetic-chromogenic, and endpoint-chromogenic methods.

Core Principle of ICH Q4B and Interchangeability

The ICH Q4B process evaluates pharmacopoeial texts to confirm scientific equivalence. When confirmed, the texts are deemed interchangeable. Q4B does not replace the compendia with a single unified chapter; it recognizes existing texts as equivalent for regulatory purposes.

Conclusion for BET

• Formal Declaration: EP 2.6.14, JP 4.01, and USP <85> are equivalent; any may be used in full for regulatory purposes in all ICH regions.

• In Practice: A manufacturer may select one complete chapter (e.g., USP <85>)—including its methods, reagents, calculations, and acceptance criteria—and have the data accepted by EMA, MHLW, and FDA.

• Conditions for Use:

  • The chosen pharmacopoeial procedure must be implemented in its entirety (no mixing of elements from different chapters).

  • Harmonization applies to the test procedures, but regional authorities may impose specific endotoxin limits or compliance conditions.
