USP <1033> provides a framework to validate performance characteristics of biological assays used to measure potency/biological activity for uses such as QC lot release, stability. While not enforceable unless referenced in a compendial requirement or regulatory filing, 〈1033〉 outlines generally accepted scientific practices for bioassay validation.
Key concepts & validation characteristics (interpreted for bioassays):
- Accuracy (trueness). Accuracy is operationally defined relative to the designated standard; it cannot be established absolutely, but can be evaluated through recovery studies using samples of known relative potency
- Precision. Because bioassays are inherently variable, precision assessment is critical at multiple levels: (the depth of precision assessment should reflect the assay’s lifecycle stage and intended use (e.g., exploratory vs. QC release))
- Repeatability (intra-assay) within a run;
- Intermediate precision (inter-assay, same lab) across days/analysts/reagent lots;
- Reproducibility (between labs) during/after transfer, when applicable.
- Specificity. Ability of the assay to selectively measure the intended biological activity of the target. Specificity should be demonstrated using orthogonal approaches (e.g., neutralizing reagents, stress-degraded or variant preparations)
- Linearity/Model suitability & Range. Demonstrate that the dose–response model used (often non-linear) appropriately describes responses over the working range, and that relative potency estimates are consistent across suitable dilutions (dilutional linearity) within that range. Define a range over which accuracy and precision criteria are met.
- Robustness/Ruggedness. Show the assay’s reliability under small, deliberate method changes (e.g., incubation time/temperature, cell density); robustness studies should be risk-based and commensurate with assay criticality; designed experiments are one option, but smaller focused studies may suffice.
System suitability & run validity. Establish prospective, quantitative run-acceptance criteria that verify assay and model assumptions before reporting potency. For relative potency methods, parallelism is both a scientific assumption of the relative potency model and a formal system suitability requirement; assays failing parallelism checks must be invalidated.
Risk-based acceptance criteria. Target criteria should balance the risks of incorrect decisions from bioassay measurements with demonstrated assay capability and, where relevant, existing specifications.
Documentation. A validation protocol defining objectives, design, acceptance criteria, materials, and analyses, and a validation report interpreting outcomes against pre-specified criteria, are expected.
